ANTIBODY TO MITOCHONDRIAL COMPLEX-I IN SOME PATIENTS WITH MULTIPLE SCLEROSIS
Authors
Abstract:
When pooled immunoglobulin G (IgG) from patients with multiple sclerosis (MS) was used to probe a human fetal spinal cord λgt 11 cDNA library, the IgG was found to bind to a predicted epitope of human mitochondrial ND4 sequence. To investigate the involvement of the ND4 as an autoantigen in MS, we determined the presence of specific antibody in the serum of MS patients and serum samples of some other autoimmune disease as controls. A peptide, which is part of the ND4 protein in human mitochondrial complex I, CysLeuAlaAsnSerAsnTyrGluArgThrHisSerArg, was conjugated with a maleimido-thiol bond to diphtheria toxoid and used as an autoantigen. To remove any IgG which bound to diphtheria toxoid and the bovine serum albumin (BSA) blocking agent in the ELISA, the sera were preadsorbed before being incubated with the conjugate. About 20% of patients with multiple sclerosis (MS) had antibody to the peptide and when present, the level was found to fluctuate. In preliminary experiments autoantibody to ND4 was found to be not specifically associated with MS. The prevalence and involvement of the autoantibody in multiple sclerosis remains to be determined.

multiple sclerosis (ms) is a demyelinating disease of the central nervous system characterized by morphological hallmarks of inflammation, demyelination and axonal loss. to date, little attention has been paid to the contribution of mitochondrial respiratory chain enzyme activities to ms. in this study, kinetic analysis of mitochondrial respiratory chain complex i enzyme (measured as nadh ferri...
full textPsychological Changes in Patients With Multiple Sclerosis
Introduction: Multiple Sclerosis (MS) as a stressful event, despite causing negative psychological changes, may lead to positive ones as well. Objective: This study was mainly conducted to examine the implications of posttraumatic growth and demoralization and also to determine the relationship between them in patients with MS. Materials and Methods: This was a cross-sectional study, in which...
full textCLONING AND SEQUENCING OF A MITOCHONDRIAL AUTOANTIGEN WITH IMMUNOGLOBULIN G FROM PATIENTS WITH MULTIPLE SCLEROSIS
Multiple Sclerosis (MS) is a chronic neurological disease of the central nervous system (CNS), characterised by a cellular immune response in early stages and demyelination of the CNS later. Although the cause of MS is unknown, there is much evidence that points to MS as an autoimmune disease. To test the hypotheses that an Autoantigen is involved in MS, we screened a ?gt11 human foetal spinal ...

Background & Aims: The prevalence of multiple sclerosis (MS) has been on the rise, especially among the youth in Iran. MS is more common in the individuals in their 20s and 40s, while women are also twice as likely to develop MS compared to men. Statistics suggest that there are 2.5 million MS patients worldwide, and this figure has been reported to be 15-30 cases per 100,000 in Iran. According...
